辣椒内生烟管菌的分离鉴定与生长条件优化

Isolation, identification and optimization of growth conditions of endophytic fungi of Bjerkandera adusta in pepper

以辣椒品种羊角椒为试验材料,从中分离筛选内生真菌,并通过形态学观察和序列分析对其进行鉴定,考察不同碳源(葡萄糖、麦芽糖、蔗糖、乳糖及可溶性淀粉)、氮源(氯化铵、硫酸铵、硝酸钾、酵母膏和蛋白胨)、促进剂(吐温-80和油酸)及pH(5.0~12.0)对该菌株生长的影响。结果表明,从羊角椒中分离出的内生真菌菌丝正面白色绒毛状,无色素产生,气生生长,质地疏松;有隔膜,担孢子椭圆形,顶端钝圆;基因序列大小为515 bp,与Bjerkandera adusta在同一个分支上,可初步鉴定为烟管菌,并命名为烟管菌LJ-1。该菌生长的最适碳源为蔗糖、最适氮源为酵母膏、最适促进剂为油酸、最适pH在6.0~8.0,上述条件下菌丝生长速度较为理想,且呈现出良好的长势状态。研究结果为辣椒内生烟管菌的深入研究提供参考。

The endophytic fungi was isolated and screened from pepper variety Yangjiaojiao, and identified by morphological observation and sequence analysis. The effects of different carbon sources (glucose, maltose, sucrose, lactose and soluble starch), nitrogen sources (ammonium chloride, ammonium sulfate, potassium nitrate, yeast extract and peptone), accelerators (Tween-80 and oleic acid), and pH (5.0-12.0) on the growth of this strain were investigated. The results showed that the front side of the endophytic fungi mycelium isolated from the pepper was white fluffy-like, no pigment production, air growth and loose texture. Septate, spore-bearing oval, apical obtuse; The gene sequence size was about 515 bp, and it was on the same branch as Bjerkandera adusta, which can be preliminarily identified as Bjerkandera adusta and named Bjerkandera adusta LJ-1. The optimal carbon source for the growth of the bacteria was sucrose, the optimal nitrogen source was yeast extract, the optimal accelerator was oleic acid, and the optimal pH was 6.0-8.0. Under the above conditions, the mycelium growth rate was ideal and showed a good growth state. The results provide a reference for further study of Bjerkandera adusta.
