The safeguarding of ecological flow in rivers and lakes is an important measure to ensure the ecological security of river basins. Based on the principles of control section selection and the characteristics of the sections, the Xinqiao Sluice was determined as the main control section for the ecological flow protection of the Niutun River basin, with the lowest ecological water level as the indicator for ecological flow protection. The target values for the lowest ecological water level at the main control section of the Niutun River were calculated using the Qp method, biological spatial method, and the lowest average flow method of the driest month, respectively, and compared with the measured water levels over the past 40 years to analyze the feasibility of the targets. The results showed that the calculated values for the lowest ecological water level at the control section using the Qp method, biological spatial method, and the lowest average flow method of the driest month were 4.60, 4.50, and 4.43 m, respectively. Considering the calculation accuracy, satisfaction level, and the strict requirements for ecological flow protection, the final target for the lowest ecological water level in the basin was set at 4.60 m, with the monthly and ten-day guarantee levels reaching 99.6% and 99.3%, respectively. Based on this, management measures and early warning measures for ecological flow protection were proposed to provide a reference for ecological protection and ecological flow safeguarding in the basin.
